There are usually a few people there but in my experience things move quite quickly. An employee there will tell you ( in Czech :) ) which forms to fill in and what you should do if you ask nicely.
If your address is not in Brno Mesto you may have to go to a different office. They may also check you residency status and you will have to register with the Financni Urad (tax office) and the health insurance office.
Good luck and keep us updated with your progress. :)

If you're non EU, you will also need to register at the business court on Husova street. They have a rather intimidating green registration form and it will cost you 5000 Koruny to register there.
It is rather unfair and discriminatory that non EU citizens still have to cough up the extra 5000 Koruny and do the extra leg work for something the Ziv office does for Czechs and EU citizens. Thankfully, unless you let your ziv list lapse, that 5000 Koruns is a one time thing.
you also don't have to register with the commercial court if you have a permanente residence permit in the czech republic no matter what passport you hold...
is it something the ziv office does for others? i have a feeling that the ziv lists of these "others" aren't registered with the commercial court at all...
http://www.mpo.cz/dokument26504.html - the english attachment that is downloadable at the bottom provide detailed information (direct download link: http://download.mpo.cz/get/30075/41186/491951/priloha004.pdf)
the first part is general information
for citizens of the eu, eea and switzerland, appendix one on pages 12-19
for persons with a permanent residence address outside of the eu, eea and switzerland appendix two in pages 20-27 (although i cannot find reference to having to register in the coomercial register, that requirement is specified in the gernaral information in the first section (page 10)
